Biological Context

Oct4 and Sox2 are transcription factors that direct development of the mammalian embryo. Recently, Yamanaka and co-workers have shown that induced pluripotent stem (iPS) cells can be generated using four transcription factors: Oct3/4, Sox2, Klf4, and c-Myc.

Oct4 consists of two POU (POU_H and POU_S) all-alpha globular domains that bind to the major groove of double-stranded DNA and are connected by a flexible linker. Sox4 is a single (HMG) domain all-alpha protein and binds to the terminal end of the DNA and also interacts with the larger (POU_S) domain.
